核质蛋白在细胞质中合成后就通过核定位信号运送到细胞核，是一种丰富的核蛋白，在核小体的装配中起作用。核质蛋白的作用在于既能促进组蛋白与DNA的相互作用形成核小体，又能避免DNA与组蛋白间因强静电吸引而形成非特异结合的不溶性聚合物，但它本身并不参与核小体的组成。
核质蛋白具有头尾两个不同的结构域，由5个单体组成，分子量为165kDa，是耐热性可溶蛋白。尾部具有核定位信号，体外实验证明只要有一个尾部结构，就可将全部的头部带入细胞核。
